I was able to make it work with the following config:
$Conf{RsyncShareName} = [
  '/home',
  '/root'
];
$Conf{BackupFilesExclude} = {
  '/home' => [
    '*/.VirtualBox/Machines',
    '*/Downloads',
    'virt_admin/Machines'
  ]
};
