It happens to all bpduplicate jobs whenever I try any DSSU Manual Relocation.

The DSSU is 3.5 TB on SATA disks, it was divided into different folder for daily, weekly and monthly backup
Hence, I have 3 DSSU setup with S:\daily; S:\weekly and S:\monthly
The DSSU is formated as NTFS

This is NOT supported - you can't put multiple DSSUs on the same disk.
