I believe so - 5.1MP6 has not supported since 6.5.3 came out so it may not even have been tested, but I expect that a remove and re-install of the current version will work just fine. Throwing it right on top of the 5.1 install probably works too but you're basically on your own.

Also, for new clients, I'm assuming I can just install 6.5.1 and upgrade to 6.5.3 (I don't need to install the 6.5.1a patch first).

We typically do the base 6.5 kit and then the 6.5.3 patch with nothing in between. No issues so far.
