Check to ensure the you can ping/DNS lookup from the "54" clients to all of 
your master and media servers.
 
Do you have a master and a seperate media server?  I have had a situation where 
the master could communicate with the client, but the client did not know about 
the media (DNS error).  Backups would start/finish if the master did not hand 
off the client.  However, if the client was "sent" to the media server, the 
backup would fail.

Hi all

2 Windows clients (out of +- 60 clients) fail with error code 54. (All other 
client backups are successful.)

The server owners insist that there is no firewall and/or security software 
installed on these machines.
