Can I somehow say don't compress anything except this one directory?

There's a workaround you can do this with. Create a second node definition for 
the client, and point it at a second dsm.opt client option file that includes 
your bad-boy directory and excludes everything else. On the server's definition 
for that node, turn compression on. For the original node's dsm.opt file, 
exclude that particular directory and include everything else. On the server's 
node definition, turn compression off.
