How do I omitt files from being backed up for one time only?

I want to run a backup of all of a hard drive, but leave out a specific
subdirectory for later backing up, as that directory has files currently
changing and I'll back them up later. If I use EXCLUDE, then that means the
directory will NEVER be backed up, where as I do want to back up the files
at some point.
